mutations in separate loci. A comparison of the present findings with those of Demerec has shown that the four phenotypic groups are identical with the four transduction classes, which suggests that each group is characterized by a block in one of the steps of tryptophan biosynthesis and that each block is produced by mutation in one gene locus. The data are therefore consistent with the following biosynthetic pathway in which each step is controlled by a single gene:

An integrated bioreactor-expanded bed adsorption system for the removal of acetate to enhance the production of alpha-interferon-2b by Escherichia coli

A stirred tank bioreactor (STB) integrated with an expanded bed adsorption (EBA) system containing anion-exchange resin (Diaion WA30) was developed for in situ removal of acetate to increase the production of α-interferon-2b (α-PrIFN-2b) by Escherichia coli (E. coli). Although the total acetate (9.79 g/L) secreted by E. coli in the integrated STB/EBA system was higher than that in a bioreactor ...

Inhibition of host protein synthesis during infection of Escherichia coli by bacteriophage T4. I. Continued synthesis of host ribonucleic acid.

The ribonucleic acid (RNA) synthesized at specified intervals during infection of Escherichia coli K-12 by bacteriophage T4 was hybridized to denatured E. coli or T4 deoxyribonucleic acids (DNA). The reactions were performed under conditions that maximized the yield and at RNA/DNA inputs such that excess DNA sites were available for all RNA species. Most of the RNA synthesized at any time durin...

Cloning and expression in Escherichia coli of DNA encoding a 60 kDa stress protein of Mycobacterium paratuberculosis, the causative agent of Johne's disease.

Polymerase chain reaction (PCR) was used to generate DNA encoding a 60 kDa stress protein of Mycobacterium paratuberculosis using primers complementary to sequences at the 5' and 3' ends of 60 kDa stress protein genes (encoding the '65 kDa antigens') of M. leprae and M. tuberculosis. The predicted PCR product of 1.8 kb contained the entire coding sequence of an M. paratuberculosis 60 kDa stress...

Isolation of the bacteriophage lambda receptor from Escherichia coli.

A factor which inactivates the phage lambda can be extracted from Escherichia coli. This factor is a protein and is located in the outer membrane of the bacterial envelope. It is found in extracts of strains which are sensitive to phage lambda, but not in extracts of strains specifically resistant to this phage. We conclude that this factor is the lambda receptor, responsible for the specific a...

Microchemostat-microbial continuous culture in a polymer-based, instrumented microbioreactor.

In a chemostat, microbial cells reach a steady state condition at which cell biomass production, substrates and the product concentrations remain constant. These features make continuous culture a unique and powerful tool for biological and physiological research. We present a polymer-based microbioreactor system integrated with optical density (OD), pH, and dissolved oxygen (DO) real-time meas...
